Overview

**3M-052-AF + alum** is a combination vaccine adjuvant composed of the synthetic small molecule TLR7/8 agonist **3M-052-AF** (an aqueous formulation of 3M-052) and aluminum hydroxide (alum). The combination is designed to enhance immune responses to co-administered antigens. *Mechanism of Action*: The primary component, **3M-052**, is an imidazoquinoline derivative that activates Toll-like receptor 7 and Toll-like receptor 8 (TLR7/8), leading to robust stimulation of innate immunity. This activation promotes strong B cell and T cell responses, including the induction of long-lived plasma cells in bone marrow and persistent antibody production[1][4]. Alum acts as a depot-forming adjuvant that helps localize antigen/adjuvant at the injection site and further enhances immunogenicity by promoting antigen uptake by antigen-presenting cells[2][5]. *Clinical Use*: This adjuvant system has been evaluated in early-phase clinical trials as part of HIV vaccine regimens using trimeric BG505 SOSIP.664 gp140 protein antigens. In phase I studies in healthy adults, it was found to be safe and well-tolerated with no serious adverse events; most side effects were mild or moderate local reactions[2][5][7].
