Alum acts as a depot-forming adjuvant that helps localize antigen/adjuvant at the injection site and further enhances immunogenicity by promoting antigen uptake by antigen-presenting cells[2][5]. *Clinical Use*: This adjuvant system has been evaluated in early-phase clinical trials as part of HIV vaccine regimens using trimeric BG505 SOSIP.664 gp140 protein antigens. In phase I studies in healthy adults, it was found to be safe and well-tolerated with no serious adverse events; most side effects were mild or moderate local reactions[2][5][7]. The regimen induced robust trimer-specific antibody responses—including neutralizing antibodies—and strong B cell/CD4+ T cell activation[2][5][7]. *Developer*: The compound was originally developed by researchers at the Vaccine Research Center (VRC), National Institute of Allergy and Infectious Diseases (NIAID), with clinical trial sponsorship from NIAID Division of AIDS/HIV Vaccine Trials Network.
