3scFv × hL0125C is a trivalent, bispecific T-cell engager developed by FrontAim Biomedicines using its molecularly grafted immunoglobulin (MG-Ig) platform. The antibody is designed to target both CD3 on T cells and Tumor-associated calcium signal transducer 2 (Trop2) on tumor cells, thereby redirecting T-cell-mediated cytotoxicity to Trop2-expressing cancer cells. It is constructed by grafting an anti-CD3 single-chain variable fragment (scFv) onto the hinge region of a full-size humanized anti-Trop2 immunoglobulin (hL0125) using a dimerization and docking domain (DDD) and anchoring domain (AD) interaction. In preclinical studies, 3scFv × hL0125C has demonstrated potent, dose-dependent cytotoxicity against various Trop2-expressing cancer cell lines, including breast cancer and colorectal cancer, and has shown stability in human plasma.
