4,11-dichloro-BBQ (DiCl-BBQ) is a small molecule member of the benzimidazoisoquinoline class that acts as a ligand for the aryl hydrocarbon receptor (AhR). It has been identified as a potent modulator of AhR-regulated transcription with potential antineoplastic activity. In preclinical studies, specifically involving hepatocellular carcinoma (HCC) cell lines such as HepG2, 4,11-dichloro-BBQ has demonstrated the ability to suppress global protein synthesis and inhibit cell growth in an AhR-dependent manner. It belongs to a category of rapidly metabolized AhR ligands (SMAhRTs - Select Modulators of AhR-regulated Transcription) designed to minimize the toxicity associated with persistent AhR activation while maintaining therapeutic efficacy in cancer and immune-mediated conditions.
