This is a combination autologous CAR T cell therapy consisting of two genetically engineered T cell products. The first component, 4-1BBζ EGFR806-EGFRt, is a second-generation chimeric antigen receptor (CAR) T cell targeting the tumor-specific epitope of epidermal growth factor receptor (EGFR) using the mAb806 binder, with a co-stimulatory domain from 4-1BB and an incorporated selection-suicide marker (EGFRt). The second component, 4-1BBζ CD19-Her2tG, is a CAR T cell targeting CD19 with a similar co-stimulatory domain and an alternative selection-suicide marker (Her2tG). This dual approach aims to target both solid tumors expressing EGFR and B cells expressing CD19 to enhance anti-tumor efficacy and persistence of the CAR T cells. Both products are manufactured from the patient's own blood-derived T cells. The therapy is under investigation for relapsed or refractory non-CNS solid tumors in pediatric and young adult populations[2][5][6].
