4-1BB (CD137) agonist antibodies are a class of immunotherapeutic agents designed to stimulate the 4-1BB receptor, a co-stimulatory member of the tumor necrosis factor receptor (TNFR) superfamily. Expressed on activated T cells and natural killer (NK) cells, 4-1BB signaling provides a potent co-stimulatory signal that enhances T-cell proliferation, survival, and effector functions, such as cytokine production and cytotoxicity. In preclinical models of multiple myeloma, 4-1BB agonists have demonstrated the ability to increase CD4+ and CD8+ T-cell infiltration in the bone marrow and spleen, thereby promoting anti-tumor immune responses. Research indicates that the therapeutic efficacy of these antibodies can be highly dependent on the specific clone and IgG isotype, with IgG2a-formatted agonists (such as clone 3H3) demonstrating superior anti-tumor activity compared to other formats in certain models. These agents are frequently investigated in combination with other immunomodulators, such as S100A9 inhibitors like tasquinimod, to mitigate the immunosuppressive tumor microenvironment and enhance overall anti-tumor immunity.
