4-1BB x NECTIN-4 bispecific antibody is a preclinical-stage immuno-oncology candidate developed by Twist Bioscience. The therapeutic is a targeted bispecific antibody designed to simultaneously bind the costimulatory receptor 4-1BB (CD137) on immune cells and the tumor-associated antigen Nectin-4 on malignant cells. Nectin-4 is frequently overexpressed in various solid tumors, including bladder, breast, and lung cancers. By tethering T cells to Nectin-4-expressing tumor cells, the antibody facilitates the localized clustering and activation of 4-1BB within the tumor microenvironment. This conditional activation mechanism is intended to enhance anti-tumor immune responses while potentially mitigating the systemic toxicities, such as hepatotoxicity, that have historically limited the clinical development of monospecific 4-1BB agonists.
