4-1BBL + IL-12 refers to experimental combination immunotherapies that co-deliver the costimulatory ligand 4-1BB ligand (4‑1BBL, TNFSF9) together with the pro‑inflammatory cytokine interleukin‑12, typically via gene therapy vectors or protein constructs, to enhance anti‑tumor T‑cell responses. Preclinical studies using adenoviral or oncolytic viral vectors encoding both IL‑12 and 4‑1BBL, or systemic administration of 4‑1BBL fusion proteins plus IL‑12 gene transfer, have shown synergistic activation of cytotoxic T cells, increased interferon‑γ production, and durable tumor regressions in mouse models of hepatic colon carcinoma and other solid tumors by simultaneously providing IL‑12–driven Th1 polarization and 4‑1BB costimulation to tumor‑reactive CD8 T cells and antigen‑presenting cells.[3][4][7]
