4-amino-2-trifluoromethyl-phenyl retinate (ATPR) is a synthetic derivative of all-trans retinoic acid (ATRA) designed to possess superior anticancer properties compared to ATRA. ATPR inhibits proliferation and promotes differentiation of acute myeloid leukaemia (AML) cells. Its mechanism involves modulation of the RARα/LDHB/ERK-glycolysis signaling axis, with a dependency on retinoic acid receptor alpha (RARα) activity and effects on glycolytic metabolism via lactate dehydrogenase B (LDHB). The compound has also demonstrated anticancer activity in models of gastric, hepatic, and breast cancer. ATPR was designed and synthesized by a team at Anhui Medical University.
