4-aminobenzoic acid hydrazide (4-ABAH) is a small molecule that acts as a potent and irreversible inhibitor of myeloperoxidase (MPO). MPO is a heme-containing peroxidase enzyme primarily expressed in neutrophils and monocytes, where it plays a critical role in the innate immune response by producing reactive oxygen species (ROS) such as hypochlorous acid (HOCl). While essential for host defense, excessive MPO activity is implicated in tissue damage across various inflammatory, cardiovascular, and neurodegenerative diseases. In oncology research, 4-ABAH is frequently used as a pharmacological tool to investigate the role of MPO-expressing innate immune cells in the tumor microenvironment. Preclinical studies have demonstrated that 4-ABAH can inhibit tumor NF-κB transcriptional activation and modulate tumor initiation and growth, effectively mimicking an MPO-null phenotype in animal models. It is primarily utilized as a research-use-only reagent in laboratory settings.
