Overview

4-CNAB is a synthetic small molecule absorption/permeation enhancer, structurally related to SNAC (sodium 8-(2-hydroxybenzamido)octanoate), developed to facilitate the oral delivery of biologics, particularly peptide and protein drugs such as insulin. Its mechanism involves transiently increasing gastrointestinal epithelial permeability, thus improving the absorption of co-administered biomolecules by forming a non-covalent hydrophobic ion-pair without reducing peptide bioactivity. 4-CNAB has been investigated in combination with insulin in clinical studies and has completed phase 1 trials for this indication[3][4][5].
