4-hydroxyderricin (4HD) is a natural chalcone derived from the Ashitaba plant (*Angelica keiskei*). It has demonstrated potential as a chemopreventive and therapeutic agent against malignant melanoma. The compound acts by directly targeting and inhibiting both BRAF (specifically the BRAFV600E mutant) and PI3-K signaling pathways. This dual inhibition blocks downstream signaling through the BRAF/MEK/ERK and PI3-K/AKT pathways, leading to G1 phase cell cycle arrest and the induction of apoptosis in melanoma cells. Preclinical studies in Pten-deficient melanoma mouse models have shown that 4HD significantly reduces tumor incidence and volume.
