4-IPP (4-iodo-6-phenylpyrimidine) is a small molecule research compound that acts as a potent, irreversible inhibitor and suicide substrate of macrophage migration inhibitory factor (MIF). It functions by covalently modifying the catalytically active N-terminal proline residue of MIF, thereby neutralizing its biological and enzymatic activities. Developed by the Centre Hospitalier Universitaire Vaudois, 4-IPP has demonstrated significant preclinical potential in oncology, specifically in inhibiting the growth and migration of osteosarcoma, lung adenocarcinoma, and colorectal cancer cells. Additionally, it shows promise in bone health by inhibiting osteoclastogenesis and promoting osteoblast differentiation. While it is widely used as a tool in MIF-related research and is more potent than the standard inhibitor ISO-1, it is currently not approved for clinical use in humans.
