Overview

4-Methoxyamphetamine (PMA) is a synthetic amphetamine derivative and designer drug with potent serotonergic activity. It acts primarily as a selective serotonin releasing agent (SSRA), increasing the release of serotonin from presynaptic neurons and inhibiting its reuptake. PMA also exhibits weak effects on dopamine and norepinephrine transporters compared to other amphetamines. Additionally, it is a potent reversible inhibitor of monoamine oxidase A (MAO-A), which can further elevate serotonin levels. The combination of these mechanisms contributes to its high toxicity profile and risk for life-threatening hyperthermia and other severe adverse effects at relatively low doses. PMA has no accepted medical use and is classified as a Schedule I controlled substance in the United States[1][2][3][6][7].
