Overview

**4-nitroquinoline-1-oxide** is a quinoline-derived synthetic small molecule, primarily used as a model chemical carcinogen and mutagen in research settings[1][5]. It induces DNA lesions, including bulky adducts, DNA strand breaks, oxidative lesions (notably 8-hydroxydeoxyguanosine), and DNA-protein crosslinks, through both direct covalent binding and the generation of reactive oxygen species (ROS) after metabolic activation[1][5][7]. Its carcinogenic activity is exploited in laboratory animals (commonly rodent oral cancer models) to mimic the effects of tobacco carcinogens and test interventions for cancer prevention and treatment[5]. The compound is not used clinically, but serves as a tool for studying DNA repair, mutagenesis, and carcinogenesis mechanisms[1][5].
