Overview

4-O-Demethylpenclomedine is a synthetic small molecule and the principal alkylating metabolite of penclomedine, a multichlorinated alpha-picoline derivative. It belongs to the polyhalopyridines class, characterized by a pyridine ring substituted with multiple halogen atoms. The compound acts as an antitumor agent through its highly reactive alkylating properties. After activation in the liver via free radical formation, it is transported to tumor cells where it forms DNA adducts and induces interstrand crosslinking through its chloro moieties. This leads to DNA damage and inhibition of cancer cell proliferation. Preclinical studies have shown activity against glioblastoma and breast tumors with lower neurotoxicity compared to penclomedine[1][3][5].
