Drug intelligence / Profile preview
4-phenylthiobutanoic acid (PTBA) is a small molecule histone deacetylase (HDAC) inhibitor that selectively targets histone deacetylase 8 (HDAC8). Discovered through a pathway-agnostic, cell proliferation-based phenotypic screen in zebrafish, PTBA has been shown to promote the proliferation of renal tubular epithelial cells (RTECs) and cardiac progenitor cells. This activity enhances tissue repair and reduces post-injury fibrosis in animal models of acute kidney injury (AKI) and cardiac injury. To overcome the permeability limitations of the parent carboxylic acid, esterified prodrugs such as UPHD25 and UPHD186 have been developed to deliver PTBA in vivo.
