4025-A (also known as SPH4025) is an investigational small molecule potassium-competitive acid blocker (P-CAB) developed by Shanghai Pharmaceuticals Holding Co., Ltd. for the treatment of digestive system diseases, specifically acid-related disorders such as reflux esophagitis. Unlike traditional proton pump inhibitors (PPIs) that require acid activation and bind irreversibly to the H+/K+-ATPase enzyme, 4025-A inhibits the proton pump in a reversible and potassium-competitive manner. This mechanism allows for a more rapid onset of action, consistent acid suppression, and improved control of nocturnal acid breakthrough compared to conventional therapies. The drug is currently in Phase 2 clinical development.
