Overview

426c.Mod.Core-C4b is an experimental HIV vaccine candidate designed as a self-assembling nanoparticle immunogen targeting the CD4 binding site (CD4bs) of the HIV-1 envelope. It is specifically engineered to elicit VRC01-class broadly neutralizing antibody (bnAb) precursor B-cell lineages, which are known for their high potency and breadth against diverse HIV strains. The immunogen consists of core inner and outer domains from the Clade C 426c transmitted founder viral envelope, with modifications such as removal of V1-V3 loops and certain glycans around the CD4bs to enhance germline binding, including retention of N276 glycan. The vaccine is adjuvanted with 3M-052-AF (a TLR7/8 agonist) plus aluminum hydroxide (Alum), aiming to boost immune responses. It is being evaluated in phase 1 clinical trials for safety and immunogenicity in both people living with HIV undergoing analytical treatment interruption and in infants perinatally exposed to HIV but uninfected[1][2][5][6][7].
