Overview

430C10 is a first-in-class, orally active small molecule FOXP3-targeting stabilizer. Identified through an AI-driven virtual screening strategy, 430C10 binds to an allosteric pocket within the forkhead (FKH) domain of FOXP3. This binding reinforces the physical interaction between FOXP3 and T-bet, thereby suppressing T-bet-driven interferon-gamma (IFN-γ) production by regulatory T cells (Tregs) and restoring Treg stability. 430C10 is being investigated for the treatment of IPEX (immunodysregulation polyendocrinopathy enteropathy X-linked) syndrome, acute colitis, and other IFN-γ-driven autoimmune and inflammatory diseases.
