Overview

43H12 is a monoclonal antibody that specifically targets Programmed Death-Ligand 1 (PD-L1). Unlike conventional PD-L1 inhibitors that primarily block the PD-L1/PD-1 interaction, 43H12 is characterized by its specific ability to block the interaction between PD-L1 and CD80 (B7-1). Research conducted at the City of Hope indicates that blocking the trans-interaction of PD-L1 on tumor cells with CD80 on T cells can augment anti-tumor immunity by promoting the expansion of CD8+ effector T cells and increasing the production of interferon-gamma (IFN-γ). This mechanism appears to be dependent on the presence of PD-1 and CD28. The antibody has demonstrated efficacy in enhancing tumor immunity in syngeneic mouse models, including B16 melanoma and MC38 colorectal cancer lines.
