441-2 is a metabolite-inspired synthetic derivative of ginger-based compounds, specifically designed to mimic the activity of ginger metabolites such as M6 and constituents like 6-shogaol. It functions as an inhibitor of phospholipase C (PLC), a key enzyme in the signaling pathways that mediate smooth muscle contraction. By inhibiting PLC, 441-2 reduces intracellular calcium levels in both airway smooth muscle (ASM) and pulmonary artery smooth muscle cells (PASMC). This mechanism leads to the relaxation of precontracted pulmonary arteries and airways, suggesting potential therapeutic utility in the treatment of vascular hypertensive disorders, such as pulmonary hypertension, as well as obstructive airway diseases. The compound is being researched by investigators at Columbia University and North Carolina Agricultural and Technical State University.
