4B07-05 is a humanized monoclonal antibody targeting **CD47** (Cluster of Differentiation 47), a transmembrane protein that functions as an innate immune checkpoint. Developed by **I-Mab Biopharma**, 4B07-05 was identified as a high-affinity variant during the humanization of the murine 4B07 clone. CD47 is widely expressed on the surface of various cancer cells and acts as a "don't eat me" signal by binding to **SIRPα** (Signal Regulatory Protein alpha) on macrophages and other myeloid cells, thereby inhibiting phagocytosis. 4B07-05 is designed to block this interaction, promoting the phagocytic clearance of malignant cells by the innate immune system. While I-Mab's lead CD47 candidate, **lemzoparlimab** (TJC4), was derived from a different clone lineage (13H3), 4B07-05 represents a distinct humanized variant within the same therapeutic program. It is characterized by its potent binding to CD47 and its potential to minimize treatment-related side effects, such as anemia, by avoiding significant binding to red blood cells.
