Overview

4C7 is a recombinant human TRAIL (Tumor Necrosis Factor-Related Apoptosis-Inducing Ligand) variant engineered for high selectivity toward the death receptor 4 (DR4, also known as TRAIL-R1). Developed by researchers at the University of Groningen, 4C7 was designed to overcome the promiscuity of wild-type TRAIL, which binds to multiple death and decoy receptors (DR4, DR5, DcR1, DcR2, and OPG). By specifically targeting DR4, 4C7 aims to induce apoptosis in cancer cells while avoiding sequestration by decoy receptors. In preclinical studies, particularly in glioblastoma multiforme (GBM) and glioblastoma stem cells, 4C7 has been investigated for its ability to trigger the extrinsic apoptotic pathway, often in combination with sensitizing agents like the proteasome inhibitor bortezomib to overcome resistance.
