Overview

4C9 is a recombinant human TRAIL (Tumor Necrosis Factor-Related Apoptosis-Inducing Ligand) variant engineered for high selectivity toward the death receptor 4 (DR4, also known as TRAIL-R1 or Tumor necrosis factor receptor superfamily member 10A). Developed by researchers at the University of Groningen, 4C9 was designed to overcome the promiscuity of wild-type TRAIL, which binds to multiple death and decoy receptors. By specifically targeting DR4, 4C9 aims to induce apoptosis more effectively in tumor cells that predominantly express this receptor, such as certain glioblastoma multiforme (GBM) cells and cancer stem cells. In preclinical studies, 4C9 has been evaluated both as a monotherapy and in combination with the proteasome inhibitor bortezomib to overcome resistance mechanisms in glioblastoma.
