Overview

4E10 (often referred to as C4E10 when produced in CHO cells for clinical use) is a broadly neutralizing monoclonal antibody (bNAb) that targets the highly conserved membrane-proximal external region (MPER) of the HIV-1 envelope glycoprotein gp41. Developed through a collaboration involving institutions like Rockefeller University and Polymun Scientific, 4E10 is one of the most broad-spectrum HIV-1 neutralizing antibodies identified, capable of neutralizing a wide array of viral isolates across different clades. Its mechanism involves binding to a specific epitope on the gp41 subunit, which is critical for the conformational changes required for viral-host cell membrane fusion. In clinical development, C4E10 was evaluated in Phase I and Phase II trials, frequently as part of a combination therapy with other bNAbs such as 2F5 and 2G12, to investigate its safety, pharmacokinetics, and potential to suppress viral rebound in HIV-infected individuals during analytical treatment interruptions.
