Overview

4ET1103 is a peripherally restricted small molecule inhibitor of mitogen-activated protein kinase interacting kinases (MNK), developed as a first-in-class treatment for neuropathic pain. The drug selectively targets MNK in the periphery to interrupt key pathways involved in pain signaling, specifically by blocking evoked MNK activity in human dorsal root ganglion neurons. This mechanism is independent of opioid pathways and aims to provide effective analgesia without central nervous system side effects or risk of addiction. Preclinical studies have demonstrated efficacy in rodent models and human neurons, with an excellent safety profile. The drug has completed IND-enabling studies and was scheduled to enter phase 1 clinical trials for neuropathic pain in 2024[1][2][3][7].
