Overview

4G8 FLT3 CAR-T is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy designed to target the Fms-like tyrosine kinase 3 (FLT3) receptor (CD135). Developed primarily at the University of Pennsylvania, this cell therapy utilizes a CAR construct featuring a single-chain variable fragment (scFv) derived from the 4G8 monoclonal antibody clone, which recognizes both wild-type FLT3 and its mutated forms, such as the internal tandem duplication (FLT3-ITD). FLT3 is frequently overexpressed or mutated in acute myeloid leukemia (AML), making it a high-priority target. Upon administration, the engineered T cells identify and bind to FLT3-expressing leukemic blasts, triggering T-cell activation, proliferation, and the release of cytotoxic granules to induce tumor cell apoptosis. It is currently being evaluated in early-phase clinical trials for patients with relapsed or refractory AML.
