4H11 CAR T cells are an investigational, adoptive cell therapy engineered from autologous T cells, genetically modified to express a chimeric antigen receptor (CAR) derived from the 4H11 antibody targeting the extracellular domain of MUC16 (also known as MUC-16ecto), a tumor-associated antigen highly specific for high-grade serous ovarian carcinoma. The therapy is further enhanced (“armored”) with a human IL-12 transgene to modulate the tumor microenvironment and an EGFRt “safety switch” that enables the targeted elimination of the infused T cells with cetuximab if required. The CAR construct contains the CD28 co-stimulatory domain and CD3zeta signaling domain, and is delivered via a gammaretroviral vector. Administration is by intravenous and/or intraperitoneal infusion. Its primary clinical investigation has been in advanced, recurrent ovarian, fallopian tube, or primary peritoneal cancers.
