Overview

**4H11CAR T cells** are investigational, genetically engineered autologous T cells expressing a second-generation chimeric antigen receptor built with the 4H11 anti-MUC16 single-chain variable fragment. The CAR recognizes the retained membrane-proximal ectodomain of mucin 16 after CA125 cleavage, commonly termed MUC16ecto or MUC16CD. Antigen binding activates CD28 and CD3-zeta signaling in the engineered T cells, driving antigen-specific cytotoxicity against MUC16-expressing tumor cells. The unarmored 4H11 CAR construct has demonstrated preclinical activity in ovarian cancer and pancreatic cancer models. The IL-12-secreting, EGFRt-containing clinical product 4H11-28z/fIL-12/EGFRt is a distinct armored derivative and is not considered synonymous with unmodified 4H11CAR T cells.
