Overview

4NSG-SLN is a nanoparticle formulation of a chemically modified gemcitabine prodrug, 4-(N)-stearoyl-gemcitabine (4NSG). Developed by researchers at Florida A&M University and collaborating institutions, it is designed to overcome the pharmacokinetic limitations of gemcitabine, such as its short half-life and susceptibility to enzymatic degradation. The 4-amino group of gemcitabine is conjugated with stearic acid to form the lipophilic prodrug 4NSG, which is then encapsulated into solid lipid nanoparticles (SLN). This formulation enhances systemic stability, bioavailability, and anticancer efficacy, particularly in pancreatic cancer models. In preclinical studies using patient-derived xenograft (PDX) models, 4NSG-SLN demonstrated superior tumor growth inhibition compared to standard gemcitabine.
