Overview

4SC-205 is an orally available small-molecule inhibitor of the kinesin spindle protein Eg5 (also known as KIF11), a motor protein essential for mitotic spindle formation and cell division. By binding to the ATP-binding site of Eg5, 4SC-205 disrupts microtubule dynamics required for proper chromosome segregation during mitosis, leading to mitotic arrest and apoptosis in cancer cells[1][6][8]. The drug is being developed primarily as an antineoplastic agent for solid tumors and lymphoma. It has shown antitumor activity in preclinical models, including neuroblastoma[3], and demonstrated safety and initial efficacy signals in Phase I clinical trials[5][8]. 4SC-205 is notable as the only orally available Eg5 inhibitor currently in clinical development worldwide[7][8]. The compound was originally discovered by Nycomed and further developed by 4SC; it is licensed to Link Health Group for development in China.
