Overview

4SCAR-276 is an autologous, fourth-generation chimeric antigen receptor (CAR) T cell therapy that targets CD276 (also known as B7-H3), a surface protein highly expressed on many tumor types but at low levels on normal tissues. The therapy uses patient-derived T cells genetically modified via lentiviral transduction to express a CAR specific for CD276, fused with an inducible apoptotic caspase 9 domain for safety. These engineered T cells are designed to recognize and kill tumor cells expressing CD276. The primary indication is refractory and/or recurrent solid tumors, including brain cancer and neuroblastoma. Developed by Shenzhen Geno-Immune Medical Institute, the drug is currently in clinical trials evaluating its safety and efficacy in patients with poor prognosis due to complex multimodal therapy resistance[1][5][8].
