4SCAR-CD44v6 is a fourth-generation chimeric antigen receptor (CAR) T cell therapy in which patient or donor T lymphocytes are genetically modified using a lentiviral vector to express a CAR targeting the CD44 variant domain 6 (CD44v6), an isoform of the cell surface adhesion molecule CD44. The engineered T cells are designed to recognize and eliminate cancer cells expressing CD44v6, which is implicated in tumorigenesis, invasion, and metastasis. This target is expressed in most squamous cell carcinomas and subsets of adenocarcinomas, melanoma, lymphoma, multiple myeloma, breast cancer, prostate cancer, and stomach cancer. The therapy aims to provide specific cytotoxicity against these cancers by redirecting the immune response toward malignant cells expressing the antigen[1][2][5][7].
