4SCAR-GD2 + 4SCAR-Her2 + 4SCAR-CD44v6 is an experimental combination cell therapy consisting of fourth-generation chimeric antigen receptor (CAR) T cells designed to simultaneously target three tumor-associated antigens: GD2, HER2 (ERBB2), and CD44v6. Developed by the Shenzhen Geno-Immune Medical Institute, this multi-targeted approach aims to address the challenge of tumor heterogeneity and prevent antigen escape, which are common causes of relapse in single-target CAR-T therapies. The "4SCAR" platform utilizes a proprietary safety-modified lentiviral vector and incorporates fourth-generation CAR constructs that typically include a CD28 or 4-1BB costimulatory domain along with additional signaling components to enhance T-cell activation, persistence, and the ability to modify the immunosuppressive tumor microenvironment. This cocktail is primarily investigated for the treatment of refractory or recurrent solid tumors, including glioblastoma, osteosarcoma, and neuroblastoma.
