Overview

4SCAR30 is a fourth-generation chimeric antigen receptor (CAR) T-cell therapy targeting CD30, developed by the Shenzhen Geno-Immune Medical Institute. It is designed for the treatment of B-cell malignancies and other hematological cancers. The 4SCAR platform utilizes autologous T cells transduced with a lentiviral vector to express a CAR construct that includes CD28, CD27, and CD3ζ intracellular signaling domains, providing enhanced T-cell activation and persistence. A key feature of this therapy is the inclusion of an inducible caspase 9 (iCasp9) safety switch, which allows for the selective elimination of the CAR-T cells in the event of severe toxicity. 4SCAR30 is often evaluated in combination with other CAR-T therapies, such as 4SCAR19, to target multiple antigens simultaneously and overcome tumor heterogeneity.
