4SCAR7U is a universal, off-the-shelf chimeric antigen receptor (CAR) T-cell therapy developed by the Shenzhen Geno-Immune Medical Institute. It specifically targets CD7, a glycoprotein expressed on the surface of T-cell and NK-cell lineages and their malignant counterparts. The "4SCAR" platform incorporates a fourth-generation CAR design featuring CD27 costimulatory signaling to enhance T-cell persistence and immune memory, alongside a built-in inducible Caspase-9 (iCasp9) safety switch to mitigate potential toxicities like cytokine release syndrome or graft-versus-host disease. As a universal product, 4SCAR7U is manufactured from healthy donor T cells, allowing for immediate availability without the need for patient-specific manufacturing. It is currently being investigated for the treatment of CD7-positive hematological malignancies, including T-cell acute lymphoblastic leukemia (T-ALL), T-cell acute lymphoblastic lymphoma (T-LBL), acute myeloid leukemia (AML), and NK cell lymphoma.
