Overview

5,6-methylenedioxy-2-aminoindane (MDAI) is a synthetic entactogen and research compound belonging to the 2-aminoindane family. Structurally related to 3,4-methylenedioxymethamphetamine (MDMA), it was originally synthesized in the 1990s by David Nichols' research group at Purdue University to investigate non-neurotoxic alternatives to MDMA. MDAI acts as a selective serotonin and norepinephrine releasing agent (SNRA), with significantly lower potency for dopamine release compared to MDMA, a profile intended to reduce dopaminergic neurotoxicity. Although it has been investigated in academic settings for its potential as a non-neurotoxic alternative to MDMA in psychotherapy, it has never undergone formal clinical development by a pharmaceutical company. Instead, it emerged on the recreational drug market around 2010 as a 'legal high' or new psychoactive substance (NPS).
