5-aminolevulinic acid hydrochloride (5-ALA HCl), known by the brand name Gliolan, is a small molecule prodrug utilized in sonodynamic therapy (SDT) for the treatment of high-grade gliomas. Administered orally, 5-ALA is preferentially metabolized within tumor cells into protoporphyrin IX (PpIX), a potent sonosensitizer. In a clinical protocol evaluated by Universität Münster, the CV01 ultrasound device is used to deliver low-intensity ultrasound to the tumor site, triggering PpIX to produce reactive oxygen species (ROS) that induce localized cell death (apoptosis). This drug-device combination is primarily investigated as a neoadjuvant treatment administered prior to surgical resection to target malignant cells in newly diagnosed patients with glioblastoma or other high-grade gliomas.
