Overview

This is a combination of two small molecule drugs, 5-aminosalicylate (also known as 5-ASA or mesalamine) and rosiglitazone. \n*5-Aminosalicylate* is an anti-inflammatory agent widely used in the treatment of mild to moderate inflammatory bowel diseases (IBD), particularly ulcerative colitis. Its mechanism involves inhibition of pro-inflammatory cytokines and mediators, with evidence suggesting it acts via activation of peroxisome proliferator–activated receptor gamma (PPARγ) in colonic epithelial cells. \n*Rosiglitazone* is a thiazolidinedione class antidiabetic drug that acts as a selective agonist for PPARγ, improving insulin sensitivity and also exerting anti-inflammatory effects by modulating nuclear factor kappa-B (NFκB) signaling. \nThe combination has been studied clinically for the treatment of mild to moderately active ulcerative colitis. Clinical trials have shown that combined therapy achieves better therapeutic outcomes than 5-ASA alone without additional side effects. The rationale for this combination lies in their shared target—PPARγ—and potential synergistic anti-inflammatory action in the colon.
