5-CNAC is a small molecule permeability enhancer developed to enable the oral administration of peptide-based therapeutics, which typically suffer from poor bioavailability. Originally part of Emisphere Technologies' Eligen platform and later developed by Novartis, 5-CNAC functions by non-covalently associating with cargo molecules like salmon calcitonin or parathyroid hormone. This association increases the lipophilicity of the peptide, facilitating its transport across the intestinal epithelium via a passive transcellular route without altering the tight junctions. 5-CNAC reached Phase 3 clinical trials as a component of oral salmon calcitonin formulations for the treatment of osteoarthritis and osteoporosis, though development by Novartis was eventually discontinued.
