Overview

5-fluorouracil + aluminum chloride hexahydrate is an investigational topical combination therapy being developed by West Virginia University for the treatment of actinic keratosis (AK). The formulation combines 5% 5-fluorouracil (5-FU), a well-established pyrimidine antimetabolite, with 15% aluminum chloride hexahydrate (ACH). 5-FU works by inhibiting thymidylate synthase, which disrupts DNA synthesis and leads to the death of rapidly proliferating dysplastic keratinocytes. Aluminum chloride hexahydrate, commonly used as a topical hemostatic agent, is included in this combination as an augmenting agent intended to enhance the therapeutic effect of 5-FU, potentially by altering the skin barrier or inflammatory response to improve lesion clearance on the scalp and forearms.
