Overview

FUFA is a combination chemotherapy regimen consisting of 5-fluorouracil (5-FU) and folinic acid (FA), also known as leucovorin. 5-fluorouracil is a fluoropyrimidine antimetabolite that acts as a thymidylate synthase (TS) inhibitor, disrupting DNA synthesis. Folinic acid is a reduced form of folic acid that stabilizes the binding of 5-FU to TS, thereby enhancing its cytotoxic activity. This specific regimen was evaluated by the University of Ulm in the FOGT5 Phase II clinical trial, which investigated the feasibility of stratifying patients with advanced or metastasized colorectal cancer based on their intratumoral thymidylate synthase mRNA expression levels to predict treatment response. FUFA represents a standard chemotherapy backbone that has been used for decades in colorectal cancer treatment.
