This is a combination regimen consisting of four agents: - **5-fluorouracil (5-FU):** A pyrimidine analog antimetabolite that inhibits thymidylate synthase, disrupting DNA synthesis and leading to cell death. - **Leucovorin (folinic acid):** Enhances the binding of 5-FU to thymidylate synthase, increasing its cytotoxicity. - **Oxaliplatin:** A platinum-based chemotherapeutic that forms DNA crosslinks and inhibits DNA replication and transcription. - **huA33:** A humanized monoclonal antibody targeting the A33 antigen, which is highly expressed in colorectal cancer cells. It binds specifically to this antigen on tumor cells, potentially enhancing immune-mediated tumor cell killing. The combination is being investigated primarily for metastatic colorectal cancer. The addition of huA33 to standard FOLFOX chemotherapy aims to improve anti-tumor efficacy by combining targeted immunotherapy with established cytotoxic agents[3].
