Overview

This is a combination chemotherapy regimen consisting of three agents: - **5-fluorouracil (5-FU):** An antimetabolite that inhibits thymidylate synthase, disrupting DNA and RNA synthesis in rapidly dividing cells. It is widely used for the treatment of colorectal and other gastrointestinal cancers. - **ICRF-159 (razoxane):** A bisdioxopiperazine derivative that acts as a topoisomerase II inhibitor and has antiangiogenic properties. It interferes with DNA repair and replication. - **Semustine (methyl-CCNU):** An alkylating nitrosourea compound that crosslinks DNA strands, leading to inhibition of DNA replication and cell death. The combination has been studied primarily in advanced colorectal cancer. Clinical trials have shown limited efficacy compared to standard regimens; no significant improvement in response rate or survival was observed over other combinations or single-agent therapies[1][2][3]. The primary toxicity associated with this regimen is hematologic suppression.
