Overview

This is a combination regimen used in oncology, primarily investigated for advanced colorectal cancer and other solid tumors. The regimen combines the antimetabolite 5-fluorouracil (which inhibits thymidylate synthase, disrupting DNA synthesis), leucovorin (folinic acid, which enhances the cytotoxic effect of 5-FU by stabilizing its binding to thymidylate synthase), and interferon alfa-2b (a cytokine that modulates immune response and may augment the effects of chemotherapy). The combination was studied to determine if double modulation of 5-FU with both leucovorin and interferon alfa-2b could improve responses and survival compared to single modulation. However, clinical trials have shown that adding interferon alfa-2b increases toxicity without improving survival[1][4].
