Overview
This topical combination formulation was developed by Massachusetts General Hospital (MGH) to serve as a control arm in clinical trials evaluating the efficacy of calcipotriol plus 5-fluorouracil (5-FU) for the treatment of actinic keratosis and the prevention of cutaneous squamous cell carcinoma. The preparation consists of 5% 5-fluorouracil cream mixed with petroleum jelly (Vaseline). In these randomized, double-blind trials, the petroleum jelly component acts as an inactive vehicle substitute for the active calcipotriol ointment used in the experimental arm, ensuring consistent application and blinding. While 5-fluorouracil is a potent antimetabolite that inhibits thymidylate synthase to disrupt DNA synthesis in proliferating tumor cells, the petroleum jelly itself is therapeutically inactive and serves primarily as an emollient vehicle.
