Overview
This topical combination consists of 5% 5-fluorouracil (5-FU) cream mixed with petroleum jelly (Vaseline). It was specifically utilized as the control preparation in a Phase 2 clinical trial (NCT02019355) conducted by Massachusetts General Hospital. The active component, 5-fluorouracil, is a pyrimidine antimetabolite that interferes with DNA synthesis by inhibiting thymidylate synthase, thereby inducing cell death in rapidly proliferating dysplastic keratinocytes. Petroleum jelly serves as an emollient and occlusive vehicle in this formulation. The trial investigated whether a combination of calcipotriol and 5-FU was superior to this 5-FU plus petroleum jelly control for the treatment of actinic keratosis and the prevention of progression to cutaneous squamous cell carcinoma, particularly in organ transplant recipients.
