5-fluorouracil + selenium is an experimental combination therapy primarily researched for the treatment of colorectal cancer. 5-fluorouracil (5-FU) is a fluorinated pyrimidine that acts as an antimetabolite, inhibiting the enzyme thymidylate synthase and interfering with DNA and RNA synthesis. Selenium is a trace element that serves as a synergistic agent; it is thought to enhance the cytotoxic effects of 5-FU by inducing reactive oxygen species (ROS)-mediated apoptosis in cancer cells while simultaneously providing a protective effect against chemotherapy-induced side effects like diarrhea and weight loss. Recent studies have utilized advanced delivery systems, such as folate-conjugated thiolated chitosan nanoparticles, to provide pH-responsive, targeted delivery to the colon, thereby increasing efficacy and reducing systemic toxicity.
