Overview
This is a combination chemotherapy regimen consisting of 5-fluorouracil (5-FU) and semustine (also known as MeCCNU). Both are small molecule antineoplastic agents. 5-fluorouracil is a pyrimidine analog that inhibits thymidylate synthase, thereby interfering with DNA synthesis and function. Semustine is an alkylating nitrosourea compound that causes cross-linking of DNA strands, leading to inhibition of DNA replication and cell death. The combination has been studied primarily in the treatment of advanced or metastatic colorectal cancer[2][3]. Clinical trials have shown modest response rates and median survival times for this regimen in patients with advanced colorectal carcinoma[2][3].
