Drug intelligence / Profile preview
5-methoxytryptamine-alpha-lipoate (MLA) is an investigational radioprotective agent and antioxidant synthesized from melatonin (5-methoxytryptamine) and alpha-lipoic acid. Originally developed by the Institute of Radiation Medicine of the Chinese Academy of Medical Sciences, MLA acts as a reactive oxygen species (ROS) scavenger to protect against radiation-induced hematopoietic injury and myelosuppression. In clinical development, MLA is formulated into CD5-targeted lipid nanoparticles (MLA@CD5LNPs) by Guangxi Medical University Cancer Hospital. This targeted formulation is designed to specifically home to exhausted T cells, reducing post-radiotherapy oxidative stress in CD8+ T cells, maintaining mitochondrial homeostasis, and restoring anti-tumor cytotoxicity and systemic immune responses in patients with nasopharyngeal carcinoma.
