Overview

5-methoxytryptophan is an endogenous tryptophan metabolite primarily produced by endothelial cells, with anti-inflammatory and anti-fibrotic activity.[1][2][3] It protects against systemic inflammation by inhibiting macrophage activation and blocking cytokine production, preserves endothelial cell function after injury by maintaining VEGFR2 activation, enhances re-endothelialization, and inhibits vascular smooth muscle cell (VSMC) proliferation and migration through suppression of p38 MAPK and NFκB signaling.[1][3] Preclinical studies show 5-methoxytryptophan attenuates the development of vascular restenosis, atherosclerosis, and fibrosis in the liver, kidney, heart, and lung.[1][3] It is investigated as a potential lead compound for pharmaceutical development against fibrotic and inflammatory diseases but is not an approved pharmaceutical.[3]
